Steps
1. Please prepare yourself: anhydrous ethanol, normal saline, centrifuge tube.
2. Remove the precipitate and wash solution as follows:
a) Washing solution A: 21 mL was added to 9 mL of absolute ethanol; 42 mL was added to 18 mL of absolute ethanol.
b) Washing solution B: 9 mL was added to 21 mL of absolute ethanol; 18 mL was added to 42 mL of absolute ethanol.
c) If the prepared precipitate and washing solution are precipitated, they can be dissolved at 37 ° C, shaken and used.
3. Take 300 μL of saliva into a centrifuge tube, add 200 μL of lysate, 20 μL of digestive juice and 4 μL of DNA Carrier, mix by shaking, and bath at 56 ° C for 10 minutes.
4. Add 800 μL of absolute ethanol and mix gently by inversion. If there is a translucent suspension, it will not affect the DNA extraction and subsequent experiments.
5. Place the adsorption column into the collection tube, transfer 650 μL of the above solution into the adsorption column, let stand for 2 minutes, centrifuge at 12,000 rpm for 4 minutes at 4 ° C, and discard the waste liquid in the collection tube;
5. Return the column to the collection tube and transfer the remaining 674 μL of the solution to the column. Repeat step 5.
6. Put the adsorption column back into the collection tube, add 500 μL of washing solution A to the adsorption column, centrifuge at 12,000 rpm for 4 minutes at 4 ° C, and discard the waste liquid in the collection tube.
7. Place the adsorption column back into the collection tube, add 500 μL of Washing Solution B to the adsorption column, centrifuge at 12,000 rpm for 4 minutes at 4 ° C, and discard the waste liquid in the collection tube.
8. Place the column back into the collection tube and centrifuge at 12,000 rpm for 4 min at 4 ° C to remove any remaining wash solution.
9. Add the adsorption column, place it in a new 1.5 mL centrifuge tube, add 30-60 μL of the eluate, let stand for 3 min, centrifuge at 12,000 rpm for 4 min at 2 °C, and collect the DNA solution. The extracted DNA can be used in the next experiment or at -20 °C.
Precautions:
Lysates, precipitates, and washings contain irritating chemicals. Take precautions during operation to avoid direct contact with the skin and prevent inhalation of nose and mouth. If you accidentally get into skin or eyes, rinse immediately with water or saline. Seek medical attention if necessary.
Storage, transportation and expiration date
This kit can be transported at room temperature, stored at room temperature (15-25 °C), valid for 12 months. Please store the digestive juice and DNA carrier at -20 °C to avoid repeated freezing and thawing.
